Product introduction
This product is a combination of quinolones, tetracycline and macrolides for the elimination of mycoplasma cells by inhibiting DNA replication and protein synthesis. Through optimization and modulation, this product not only has low cytotoxicity and fast mycoplasma removal rate, but can completely remove mycoplasma contained in cultured cells within about 7 days. After normal medium is replaced, the cells are in good condition, and there is no repositive phenomenon in continuous multi-generation culture.
Storage and transportation

steps
1. Preparation medium A: 1 mL SweMyco Elimination reagent-1 (100×) was added to 100 mL cell culture medium (containing serum, but excluding double antibodies), and thoroughly mixed;
2. Preparation medium B: 1 mL SweMyco Elimination reagent-2 (100×) was added to 100 mL cell culture medium (containing serum, but excluding double antibodies), and thoroughly mixed;
3. Culture medium A was used for routine cell culture for 4 days when mycoplasma infected cells were subcultured or replaced with fluid; Then medium B was replaced for routine cell culture for 3 days.
4. After one round of culture (7 days), some cells were cultured in 6-well plates (or other culture vessels) and replaced with complete medium without SweMyco Elimination Reagent. The remaining cells were cultured alternately in medium A and B.
5. Cells in 6-well plates were cultured in SweMyco Elimination Reagent medium for 2-3 days, and then tested for mycoplasma when the confluence of cells reached more than 90% (DIB1915, DIB1916, DIB1917);
6. Perform the following operations based on the test results:
A) Positive results: Repeat steps 3-5 until the test results are negative.
B) Negative results: The cell group containing SweMyco Elimination Reagent medium was replaced with normal complete culture medium. After a period of culture, mycoplasma test was performed. Negative results of retest indicated completion of Elimination.
Matters needing attention
1. The product has been tested in a variety of mycoplasma infected cell lines, but different cell lines have different sensitivity to the reagent. If the toxicity to the cell is too great or the removal effect is not good, the concentration of the product can be adjusted appropriately or the removal time can be extended.
2. There are many ways for cells to be infected with mycoplasma, so it is recommended to add mycoplasma prevention reagent (DIB4024 is recommended) in daily culture.
3. For cells that have been determined to be contaminated with Mycoplasma, try to use a separate ultra-clean table during operation to avoid cross-contamination.
